چکیده

Piezoelectric materials such as quartz can be used to detect mass change due to their piezoelectric properties. Quartz Crystal Microbalance (QCM) is an extremely sensitive mass sen frequency biosensor mass mea for QCM DNA biosensor was developed. The potential for QCM to be employed as immunosensor sor, capable of measuring sub-nanogram levels by detecting change in the of a quartz crystal resonator, which makes it a promising candidate for applications. In this project, the mechanistic theories related to QCM mass measurement was reviewed and summarized. Based on the theories, a lab-scale QCM surement unit was set up. Fundamental theories and experimental techniques DNA detection were investigated, and a protocol for the construction of QCM

Oligonucleotide quartz crystal microbalance sensor for the microalgae Alexandrium minutum (Dinophyceae).

We report the immobilization on a gold surface of a 20-base DNA probe labeled with disulfide group and on the selective hybridization with the complementary 20-base DNA strand. The oligonucleotide probe is the complementary strand of a partial sequence of the gene encoding for a large ribosomal RNA sub-unit which is a coding sequence of Alexandrium minutum DNA, a microalgae that produces neurot...

Microgravimetric DNA sensor based on quartz crystal microbalance: comparison of oligonucleotide immobilization methods and the application in genetic diagnosis.

We report on the study of immobilization DNA probes onto quartz crystal oscillators by self-assembly technique to form variety types of mono- and multi-layered sensing films towards the realization of DNA diagnostic devices. A 18-mer DNA probe complementary to the site of genetic beta-thalassaemia mutations was immobilized on the electrodes of QCM by covalent bonding or electrostatic adsorption...

A Quartz Crystal Microbalance (QCM) Study of Single-Strand DNA Hybridization and Hydrolytic Cleavage

The use of a quartz crystal microbalance (QCM) for monitorin g in situ the immobilization of singlestrand DNA marked with mercaptol group at the 5'-end on the surface of a gold-filled 7.995 MHz AT-cut quartz crystal by Au–S bond with the self-assembly technique is reported. The hybridization of ssDNA with complementary 10-mer ODN and 8-mer ODN is described. The QCM was also employed to analyze ...

In situ QCM DNA-biosensor probe modification

We report on a probe modification of a quartz crystal microbalance (QCM) DNA-biosensor that permits to reversibly change the DNA sequence detected. A QCM DNA-biosensor was designed by immobilization of a 20-base DNA-disulfide probe on the gold-covered quartz surface of a 27 MHz microbalance (9 MHz, third overtone). After immobilization on the gold covered quartz surface, this probe was modified...

Surface plasmon resonance spectroscopy and quartz crystal microbalance study of MutS binding with single thymine-guanine mismatched DNA.

MutS is a DNA mismatch binding protein that recognizes heteroduplex DNA containing mispaired or unpaired bases. In this study, we employ a quartz crystal microbalance (QCM) and a surface plasmon resonance (SPR) device for the study of MutS binding with DNA containing a single Thymine-Guanine (T-G) mismatch at different sites. Multi-step surface binding reactions are involved in the study, inclu...
